为了解华东地区猫泛白细胞减少症病毒(Feline Panleukopenia Virus,FPV)的分子遗传变异规律和分子流行特征。本研究采集了华东地区28份猫泛白细胞减少症病料,扩增VP2基因全长,进行序列分析。结果显示28株之间的同源性为98.9%-100%,与疫苗株(CU-4株)的同源性为99.02%-99.54%;系统发育分析显示有6株与疫苗株位于同一分支,而其余22株在另一分支;关键氨基酸位点分析显示有22株VP2蛋白的91位点存在由A到S的突变;VP2蛋白与转铁蛋白受体(Transferrin receptor,TfR)分子对接显示TfR/FPV-VP2-91S复合物形成过程中的氢键和盐桥作用明显多于TfR/FPV-VP2-91A复合物的形成过程,表明VP2蛋白第91位点突变为S后与TfR结合力增强;分子流行特征分析显示2017-2023年中国FPV VP2蛋白第91位点突变率逐年上升,除中国南部地区未出现91S突变株外,中国其他地区均存在91S突变株,且在我国华东地区最为明显,占比达71.95%,FPV VP2蛋白第91位点突变为S已经成为华东地区FPV流行毒株的主要分子特征。本研究结果丰富华东地区FPV的分子流行病学调查,为探索诊断、预防和治疗猫泛白细胞减少症的新策略提供理论基础和关键靶点。
Abstract:In order to understand the molecular genetic variation of Feline Panleukopenia Virus (FPV) in East China. In this study, 28 samples of suspected cat panleukopenia in East China were collected, and the full-length VP2 genes of 28 strains were amplified and sequenced. The results showed that the homology of the 28 FPV VP2 genes was 98.9% -100%, and the homology with the vaccine strain (CU-4) was 99.02% -99.54%; phylogenetic analysis showed that 5 of the 28 FPV VP2 genes were located in the same branch with the vaccine strain, while the remaining 23 strains were distant from the vaccine strain. Key amino acid analysis showed that there was a mutation from A to S at the 91 site of VP2 protein in 22 strains. The molecular docking of VP2 protein and transferrin receptor (Transferrin receptor, TfR) showed that the hydrogen bonds and salt bridges in the formation of TfR/FPV-VP2-91S complex were significantly more than those in the formation of TfR/FPV-VP2-91A complex, indicating that the binding force of VP2 protein to TfR was enhanced after the 91 st site was mutated to S; the molecular epidemiological characteristics showed that the mutation rate of the 91st site of FPV VP2 protein in China increased year by year from 2017 to 2023.Except for the absence of 91S mutant strains in southern China, there were 91S mutant strains in other regions of China, and the most obvious in East China, accounting for 71.95%. The 91st site of FPV VP2 protein was S, which has become the main molecular feature of FPV epidemic strains in East China.This study enriches the molecular epidemiological investigation of FPV in East China, and provides theoretical basis and drug targets for exploring new strategies for the diagnosis, prevention and treatment of immunosuppressive diseases.
