为了探讨一氧化氮(NO)对马鹿精子获能及顶体反应(AR)的影响,本试验以塔里木马鹿冻精为试验材料,随机取出塔里木马鹿冻融精子,在含四种不同浓度(10、50、150、300nmol/L)的NO的SP-TALP获能液中进行5个不同时间段(1、2、4、6、8h)的获能培养,采用金霉素(CTC)染色法和显微镜观察法来评价精子活力、超激活运动力(HAM)、精子获能率、顶体反应率等各项精子体外获能指标,探讨一氧化氮对马鹿精子活力、存活时间、精子获能率及顶体反应率的影响。试验结果表明,低浓度(10nmol/L)NO较好地维持马鹿精子活力及超激活运动力,精子获能水平有显著地提高(P < 0.05);而高浓度(150—300nmol/L)的NO显著抑制马鹿精子活力,HAM及其获能和顶体反应(P < 0.05)。以上结果提示NO对马鹿精子体外获能具有双重性,低浓度的NO对精子获能促进作用、而高浓度的NO对精子获能抑制作用。
Abstract:In order to investigate the effect of nitric oxide (no) on sperm capacitation and acrosome reaction (AR) of red deer, the frozen semen of Tali Trojan deer was taken out randomly, and the capacitation culture was (1, 2, 4, 6, 8 h), Aureomycin (CTC) staining and microscopic observation were carried out in sp-talp capacitation solution containing four different concentrations of no (10, 50, 150, 300 nm) for five different periods of time used to evaluate various sperm capacitation indexes in vitro, such as sperm motility, hyperactivated motility (HAM), sperm capacitation rate and acrosome reaction rate, and to explore the effects of nitric oxide on sperm motility, survival time, sperm capacitation rate and acrosome reaction rate of red deer.The results showed that low concentration (10nm) no could maintain the sperm motility and hyperactive motility of red deer, and the sperm capacitation level was significantly increased (P < 0.05); High concentration of no (150-300 nm) significantly inhibited the sperm motility, ham, capacitation and acrosome reaction of red deer (P < 0.05). The above results suggest that no has dual effects on in vitro capacitation of red deer sperm. Low concentration of no promotes sperm capacitation, while high concentration of NO inhibits sperm capacitation.
