梅花鹿茸生长过程中顶端茸皮组织HDGF基因表达差异分析

摘要:

为了探究与细胞增殖、分化密切相关的肝癌衍生生长因子(Hepatoma-derived growth factor,HDGF)基因在梅花鹿茸快速生长过程中的作用,分析其在茸皮组织不同生长时期的表达差异。以梅花鹿茸生长前、中和后期的顶端茸皮组织为材料,通过克隆获得HDGF基因cDNA的完整编码区序列,对其编码蛋白的结构特征与理化性质进行分析。利用实时荧光定量PCR技术检测在三个不同生长时期的相对表达水平。结果显示,成功克隆获得梅花鹿HDGF基因,全长为720bp,共编码239个氨基酸;蛋白质相对分子量为26534.94;理论等电点pI值为4.70;生物信息学分析显示,该蛋白为不具有信号肽、能稳定存在的亲水性蛋白,二级结构以无规则卷曲(56.07%)为主;进化树分析显示其与马鹿亲缘关系最近,相似性高达99%。基因在梅花鹿茸皮组织的各生长时期均有表达,且存在差异。前期与中期的表达量差异不显著(P>0.05),前期与后期的表达量差异极显著(P<0.01),中期与后期的表达量差异显著(P<0.05)。综上,HDGF基因编码蛋白是在进化上保守的核蛋白,其mRNA在梅花鹿茸茸皮组织中的表达量随生长期推进而显著增加,推测该基因通过促进细胞增殖等功能,在鹿茸快速生长中发挥重要的调控作用。

Abstract:

In quest of elucidation regarding the functionary aspect of the hepatocellular carcinoma-derived growth factor (HDGF) gene, entangled intrinsically with cellular proliferative and differentiative processes, upon the brisk expansion of sika deer antlers, an evaluation was conducted on expression variances manifested within velvet antler dermal tissues across discrete developmental phases. Apical samples from said tissue, extracted comprehensively at incipient, medial, and terminal stages of antler maturation in sika deer, bore utility as foundational experimental objects.The complete coding sequence (CDS) of the HDGF gene cDNA was obtained through cloning, and the structural characteristics and physicochemical properties of its encoded protein were analyzed. Detectable levels of HDGF's relative transcript abundance amid three distinctive stages unfurled via accurate real-time quantitative PCR proceeded to documentation.Observational outcomes demonstrated triumphant cloning of the sika deer's HDGF gene encompassing a length of 720 base pairs encoding precisely 239 genetic kinship with red deer , intimacy reaching 99% likeness. Pattern recognition disclosed HDGF transcripts distributed consistently within velvet antler epidermis adjoined through all examined phases, albeit markedly variable. Expressive discrepancies between initial versus intermediate stages lacked statistical gravitas (P>0.05); contrastively, divergence marking initial and concluding epochs displayed profound significance conducive (P<0.01). Midway and terminal stage differentiation resonated disparity (P<0.05).Final deductions propose that the characterized HDGF encapsulates an evolutionarily preserved nuclear protein whose mRNA presence steadily intensifies progressively augmenting throughout advancing sika deer antler development periods. Inferred tentatively stands its pivotal involvement in orchestrating robust antler proliferation perhaps coadjutant to cellular division and auxiliary functional dynamics.
